How a master volume type amp responds to delay going straight in verses through teh amps effects loop. An amps preamp stage is essentially an overdrive pedal, so running your whole board into the front of a vox type amp could potentially be a muddy mess. Effects loops are designed for time based effects such as delays and reverbs to act upon your drive signal rather than becoming your drive signal. So you will get a significantly clearer and more dynamic tone using time based effects through your effects loop rather than running everything preamp. some amps that have 'high headroom' distort later than others (fenders are known for this) so its not as necessary to use an effects loop on an amp like that. again, it all comes down to preference and there is never a right or wrong way to do things.
